The Issue We Refused to Acknowledge
India has been selling probiotics—live bacteria marketed for digestive health—since the late 1990s. By 2024, the Indian probiotic market had crossed ₹1,500 crore. Yet, cases of IBS continued to rise. Bloating, acidity, and GERD had become almost routine in urban lifestyles. Antacids were being prescribed on an enormous scale, and still, patients kept coming back to clinics with the same issues—even after six months.
The clinical frustration was undeniable. Probiotics were prescribed. Antacids were given. Dietary changes were recommended. Patients returned after six months—only for the same cycle to repeat itself. Over time, it became clear that the approach, not the patients, was the problem.
The probiotic market thrived. The gut health crisis deepened. These two things were true at the same time because the wrong problem was being solved.
Why Third Biome, Why Chennai, Why Now
Third Biome was founded in Chennai by Jayavardhini and her co-founder, a physician. The founding team brings together expertise in pharmacology, research and development, gut health, and biomedical business strategy. Their advisory board includes functional medicine practitioners and academic pharmacologists.
Chennai was chosen intentionally. South India has a high concentration of health-conscious, digitally active, middle-income urban professionals. The Ingredient: Thirdbiome GTB (Microencapsulated Glycerol Tributyrate)
The Third Biome's central hero chemical is not a strain of bacteria. Thirdbiome GTB is a molecule that functions as a precise butyrate delivery vehicle.
It is a naturally occurring ester of glycerol and butyric acid. Pharmaceutical literature has extensively reported the formulation issue with raw Thirdbiome GTB: it is volatile, has an unpleasant smell, and is absorbed too quickly in the upper gut before reaching the colon, where it is required. This issue was fixed by Third Biome's R&D using their proprietary microencapsulation technology, which consists of protective microspheres that withstand stomach acid, release butyrate at intestinal pH, and transport it straight to the colonocytes, or gut lining cells, that require it most.
We refer to this as Thirdbiome GTB (microencapsulated glycerol tributyrate). What We Built: The T3B Club and the Cohort Model
We conducted four cohorts of the T3B Club, our membership-based, physician-led 30-day postbiotic program, prior to Third Biome releasing a public product. More than 300 individuals from Chennai, Bangalore, Hyderabad, Coimbatore, and Kochi have completed at least one cohort. This was not a promotional activity. Our go-to-market approach included clinical validation.
The model of the 30-day cohort was intentional. Every three to five days, the lining of the gut regenerates.
